In hop (Humulus lupulus L.), prenylated chalcones, like xanthohumol and desmethylxanthohumol, have received considerable attention due to their anti-cancer properties. However, their concentration in wildtype plants is rather low. To enhance their production, it would be interesting to modify the regulation of genes involved in the biosynthesis of flavonoids. Towards this goal, the endogenous regulatory gene hlmyb3 was genetically introduced into hop cultivar 'Tettnanger' by Agrobacterium-mediated genetic transformation. The endogenous hlmyb3 was stably incorporated and expressed in the hop genome. By quantitative real-time PCR, it was demonstrated that hlmyb3 was highly expressed in transgenic plants compared to wildtype plants. Moreover, transgenic plants with elevated expression of chs_h1, chi, f3'h, vps, and omt1 were identified.
