The ornamental nursery industry of Atlantic Canada, Memorial University of Newfoundland Botanical Garden (St. John's) and Nova Scotia Agriculture College (Truro) have initiated PlantAtlantic research and development programme to increase the palette of ornamental plant material available to the region and to stimulate their home-based production. It focused on plant breeding, selection, tissue culture, biotechnology, production research, commercialization and marketing for the horticulture industry. Sources included heirloom and hybrid varieties from plant collections throughout the region and the development of new varieties through breeding programmes. This five year (2004-09) initiative made an important contri-bution to the technical and market competitiveness of ornamental plants in Atlantic Canada. The research is already yielding revenue from plant sales and royalties. Over 25 tissue culture protocols have been developed including these for a new mock orange hybrid, Philadelphus 'Starbright', Sorbaronia - a hybrid between Sorbus and Aronia, and the Swedish Columnar Aspen, Populus tremula erecta. Aseptic cultures were established using shoot tips and meristems as explants. Media containing various concentrations of plant growth regulators (PGR's) were tested in multiplication and rooting. The factorial design indicated significant (p<0.0001) interaction between PGR's. Statistically, MS medium supplemented with 0.1 mg L-1 of IBA and 0.5-1.0 mg L-1 of BAP proved the most effective rapid multiplication of Sorbaronia and other cultures. Regeneration protocols from leaf discs, basal leaf/petiole and internode explants for transformed tissues have been developed for 'Starbright' and Swedish Columnar Aspen. The gene encoding green fluorescent protein (GFP) was used as a marker to label these two species by using Agrobacterium mediated transformation into cells. The marker GFP was confirmed in Swedish Columnar Aspen transformed plants grown in vivo by using a low-light fluorescent imaging.
