Genome identification of triploid loquats by genome in situ hybridization technique.

Genome in situ hybridization technique (GISH) was improved and applied to identify genomes of some triploid seedlings of loquats (Eriobotrya japonica) produced from tetraploid x diploid crosses. Parental DNA was used as probes and blocking DNA. The results indicated that the source of hybrid somatic chromosomes can be accurately distinguished with genome DNA of one parent as a probe, and that of the other parent as blocking DNA at 40 x concentration. The results of one analysis showed that 17 chromosomes came from the male parent, and 34 from the maternal parent. No chromosome structure variation such as interchange and inversion were found.
