In Chinese cymbidium, in vitro micropropagation usually utilizes rhizomes as explants. We cultured rhizomes of Chinese cymbidiums including Cymbidium sinense, C. formosanum and C. faberi in liquid media in order to induce multiple shoot proliferation and plant regeneration. The application of naphthalene acetic acid (NAA) to the liquid medium had a positive effect on rhizome proliferation. We used Murashige and Skoog (MS) basal medium with cytokinins at different concentrations to compare their effects on the number and the rate of shoots regenerated on the rhizomes. Shoot formation was not observed in C. formosanum and C. faberi. In C. faberi, nodular-liked structures were observed that differentiated into shoots after prolonged culture. In C. sinense, shoots differentiated from rhizome sections in liquid medium containing 6-benzyladenine (BA) and/or Thidiazuron (TDZ). Shoot differentiation was enhanced when supplemented with TDZ in contrast to BA. However, the growth of shoots was better in the BA containing medium. This research offers a new path of in vitro mass proliferation of shoots from rhizomes in Chinese cymbidiums.
