The state of DNA methylation in roots and shoots of sunflower (Helianthus annuus L.) 5-day old seedlings, which essentially differ in amounts of tissue-specific genes were compared to reveal the role of enzymatic DNA methylation in the regulation of sunflower genome expression. Using methyl-sensitive enzymes of restriction we defined organ-specificity of methylation of CCGG-sequences of DNA. External cytosines of CCG -- trinucleotide are methylated in roots, where less diverse genes are expressed while this ones in shoots are undermethylated. The functional activity of structural genes of seedlings is inversely coordinated with DNA methylation level. We assume that demethylation of CCGG of DNA palindromic sites may be associated with the transcription of group of tissue-specific genes of shoots.
