Anti-ulcer Effects of Ag * Zn-Zeolite in Rats

The presnt study was designed to clarify the effects of Ag * Zn-Zeolite, Ag-Zeolite, Zn-Zeolite and Zeolite on ethanol- or restraint plus water immersion stress-induced gastric mucosal injury and the healing of acetic acid-induced gastric ulcers in rats. Oral administration of Ag * Zn-Zeolite markedly prevented the ethanol-induced (37.5 - 150 mg/kg) or stress-induced gastric mucosal injury (150 and 300 mg/kg). Repeated oral administration of Ag * Zn-Zeolite (37.5 - 150 mg/kg twice daily) accelerated the healing of acetic acid-induced gastric ulcers in a dose-dependent manner. Among tested compounds, Ag * Zn-Zeolite was the most effective in these three models. Ag-Zeolite failed to promote the gastric ulcer healing. Zeolite was ineffective in preventing the stress-induced gastric mucosal injury and in promoting the gastric ulcer healing. Ag * Zn-Zeolite (37.5 - 150 mg/kg, p.o.) dose-dependently increased the gastric mucus content, although this compound failed to inhibit the basal gastric acid secretion. In addition, Ag * Zn-Zeolite showed the most potent superoxide anion (O_2~-)-scavenging action in vitro. These results indicate that Ag * Zn-Zeolite has the most potent anti-ulcer effect. The gastric mucus-increasing and O_2~--scavenging actions of Ag * Zn-Zeolite may be, at least in part, related to the anti-ulcer effect of this compound.
