ELISAs for detecting vitellogenin in the fathead minnow(Pimephales promelas)-a critical analysis.Response to Mylchreest et al.,Comp Biochem Physiol C 134:251-257,2003

A paper published in Comparative Biochemistry and Physiology Part C (2003),251-257,by Mylchreest et al.(2003),compared the ability of a homologous fathead minnow (Pimephales promelas) vitellogenin (fhm-VTG) ELISA and a carp (Cyprinus carpio) vitellogenin (c-VTG) ELISA to detect and quantify fathead minnow vitellogenin.In that paper,a number of statements were made that were either incorrect and/or unsubstantiated leading,we believe,to an inaccurate portrayal of assay performances for the VTG ELISAs studied.Vitellogenin is a key biomarker for exposure to oestrogenic chemicals and may emerge as an endpoint in OECD and USEPA test guidelines for endocrine disrupting chemicals.It is therefore important that the assays available for quantifying vitellogenin are critically evaluated.Equally,however,the merits and limitations of the different assays available need to be reported accurately.Here we provide a critical analysis of the assay performances of the fhm-VTG ELISA and c-VTG ELISAs for detecting fhm-VTG.
