Somatic hybrid were produced by symmetrical fusion between dihaploid clones derived from Solatium tuberosum L. cv. May Queen and the dihaploid wild species S. etuberosum. Mesophyll protoplasts of may Queen dihaploid clones were treated with iodoacetamide and electrically fused with mesophyll protoplasts of .S. ctiibcrosum clone W091. The putative hybrid calli were green in color and easily differentiated from the yellow calli of W091. These putative hybrid calli were selected and cultured in the shoot differentiation medium. Regenerated plants were cultivated in a glasshouse and identified as somatic hybrids based on their morphological characteristics and RFLP analysis using the glutamine synthetase cDNA clone as a probe. The ploidy level of the hybrids was analyzed with a flow cytometer. Of the 28 hybrids analyzed, 10 (35.7%) were 4x and morphologically normal, 4 (14.3%) were 6x, 6 (21.4%) were 8x and 8 (28.6%) were mixoploid.
