It has been reported that willow trees contain a significant amount of salicin, which is an analogue of acetylsalicyclic acid (ASA or aspirin). Several analytical methods have been attempted to quantitate this natural product. However, there have been no reports for the quantitative analysis of salicin by ATR-FT-IR spectrometer. In this study, a crude salicin mixture was extracted from barks and leaves of a willow tree, Salix Babylonica, and was quantitatively characterized by a ATR-FT-IR spectrometer. A quantitative standard salicin curve with calculated infrared absorbances of characteristic peaks between 1000 cm~(-1) and 1250 cm~(-1) was successfully obtained with a statistically excellent correlation coefficient (r~2=0.9922). The obtained standard salicin quantitation curve was then used to estimate the salicin amount in the willow tree crude extract with reproducible infrared absorbance of the extract, giving the concentration of 2.1% (±0.1) for the salicin amount in the willow tree crude extract.
