Ototoxicity may occur after administration of various clinically important chemotherapeutic cancer agents. Depending on the mechanisms of action and dosages of the specific agents, hearing loss may be temporary or permanent. Nitrogen mustards are well known ototoxic drags. In 1968, Cummings described a loss of hair cells of the organ of Corti in an animal model (cat) after intravenous administration of nitrogen mustard; temporal bone histopatho-logy demonstrated cochlear hair cell loss. High doses (0.6-1.5 mg/kg) of chlormethine (mechlor-ethamine), a nitrogen mustard, cause severe irreversible hearing loss.
