Seeds from undehised green capsule of Cymbidium aloifolium (L.) Sw. were sown in vitro and germinated asymbiotically to produce protocorm-like bodies (PLBs) within 28 days. Seed germination and PLB development were more rapid on the Knudson C basal medium when compared to MS basal medium. Knudson C medium containing 15% coconut water was effective for high percentage (80.17%) of plantlet development from PLB when cultured in liquid medium on filter paper bridge compared to solid medium (48.37%). Secondary PLBs were obtained during subculturing on the same Knudson C medium. Addition of 15% (v/v) coconut water was most effective for PLBs proliferation and plantlets development. Rooting on the young shoots was induced in Knudson C medium fortified with1 mg IBA. The plantlets with well-developed shoots and roots were transferred to community pots in greenhouse with 90% survival. The proposed protocol can be used for the rapid and efficient mass propagation and conservation of this orchid because it isvery much cost effective, reproducible and reliable.
