The effect of Macrophomina phaseolina infection was evaluated under in vivo and in vitro conditions in four chickpea cultivars, in relation to the variation in level of salicylic acid, phenylalanine ammonia-lyase and proteins within the host. Analysis of plants after every 24 hours of infection showed increased levels in defense related compounds to varying degrees at different stages of infection. They increased prior to symptom development and declined with the death of the infected tissues, but the concentrations were higher at every stage of disease development relative to healthy tissues. A similar trend of increased levels of defense compounds was observed under both in vivo and in vitro conditions. The production of defense related molecules was relatively less in excised plants in comparison to intact plants. Significant differences were encountered among infected plants depending on experimental conditions and cultivars used. The results showed that defense related molecules accumulated more in RSG-896 in comparison to other cultivars of chickpea. Differences in SDS-PAGE band profiling of total proteins were also observed in cultivars due to infection with pathogen. Salicylic acid accumulation and leading to enhanced phenylalanine ammonia lyase activity along with protein content in the compatible host-pathogen combination presumptions that salicylic acid and phenylalanine ammonia lyase participated in the chickpea resistance to M. phaseolina infection.
