Milky mushroom (Calocybe indica) is a new addition to the domestic mushroom family suitable for tropical region, next to paddy straw mushroom. The experiments were conducted to explore the role of different temperatures, pH levels and media on growth of milky mushroom and yield attributes for identification of a potential strain. In this study, fluffy growth appeared in strains CI-3, CI-4, CI-5 and CI-10 while cottony appearance was exhibited by CI-6. The colonies of strains CI-3 and CI-10 were observed as creamy white, while strains, CI-4, CI-5 and CI-6 were white. The circular colonies were observed in strains, CI-3, CI-4, CI-6 and CI-10. However, the colony of strain CI-5 was observed as irregular in shape. The breadth of pileus was significantly broader in strain CI-6 (5.3 cm) and narrower in strain CI-10 (3.7 cm). Maximum pileus weight (25 g/fruit body (FB)) and stipe length were observed in strain CI-6 followed by CI-4 (23 g/FB), CI-10 (22 g/FB) and CI-6 (21 g/FB). All strains showed more mycelial growth at 28 C temp. and 7.5 pH on 8 d after inoculation. At 28 C and 7.5 pH, strain CI-6 showed maximum mycelium growth (9 cm) while other strains showed significant variation diametric growth from 8-9 cm. Maximum radial growth (9 cm) was found in malt extract agar medium followed by potato dextrose agar medium and minimum was in sugarcane bagasse extract agar medium. Strain CI-6 took a minimum period for spawn run (16 d), pinhead formation (16 d) and for the first harvest (24 d). More pinheads were observed in strain CI-6 (64/bag) with its highest yield (620 g/kg of dry substrate with 62% BE). Average weight per sporophore was also high in strain CI-6 (32 g).
