Immuno diagnosis of Tomato leaf curl virus (ToLCV) from Tamil Nadu.

Tomato leaf curl virus (ToLCV) a begomo virus, is one of the major limiting factors in tomato growing regions of India. The triple antibody sandwich enzyme linked immunosorbent assay (ELISA) was adopted for the detection of tomato leaf curl disease using commercial polyclonal and monoclonal antibodies specific to Indian Cassava Mosaic Virus (ICMV) and African Cassava Mosaic Virus (ACMV). In the ELISA assay, the samples of leaf curl affected tomato leaves gave a different epitope profile, which resulted in strong reactions with specific monoclonal antibodies (MAb) (SCR 18, SCR 55 and SCR 62) and moderate reactions with ACMV (SCR 12, 17, 20, 21, 22, 23 and 32). ToLCV (Tamil Nadu) had the shape typical for geminivirus measuring c.30x20 nm and contained a single major protein of estimated mol.wt. c.35,000. They reacted in immunosorbent electron microscopy with MAb to ACMV SCR 18. ToLCV reacted with eight of the 14 MAbs specific for different epitopes in the particle protein of ACMV and two of the five MAbs specific to particle protein of ICMV.
