Development of Genus Specific rDNA Based Marker for Detection of Trichoderma Species

The genus Trichoderma includes biocontrol agents (BCAs) effective against soil-borne plant pathogenic fungi. Several potentially useful strains for biological control are difficult to distinguish from each other and from other fungal species in the field. So, there is a need to design and validate oligonucleotide genus specific primers for amplifying the predominant members of Trichoderma species using polymerase chain reaction. Therefore, an oligonucleotide primer set (TvPF and TvPR) based on internal transcribed spacer (ITS) regions of ribosomal DNA was designed and tested. This primer gives 245bp amplification only with Trichoderma genus. Primers were validated against other genus like Aspergillus sp., Chetomium sp., Fusarium sp. and Pestalotia sp.
