Identification of Bioactive Molecules in Ganoderma lucidum

Ganoderma lucidum is one of the well-known mushrooms used in medical and therapeutic practices. We made nutritional and physiological studies to obtain optimum mycelial biomass. A maximum growth was observed in a complete yeast extract medium containing glucose and sodium nitrate as carbon and nitrogen sources, respectively. The physiological studies revealed pH 6.0 and 25 C as optimum for fungal growth. The bioactive compounds from G. lucidum extracted in ethanol were subjected to high performance liquid chromatography (HPLCusing Shimadzu LC10 with a high rate flow of water and acetonitrile (80:20) at 1.0 ml/min through C-18 reverse phase column. Detection was carried out by photodiode array detector at 195 nm. The bioactive compounds from myceliumand fruit body were extracted after treating them with 80% ethanol, 1% ammonium oxalate, 5% sodium hydroxide and glacial acetic acid. The compounds identified in mycelium were beta(l-3)-D beta(l-6)-D glucan and a glycoprotein. The fruit body showed thepresence of a glycoprotein, beta(l-3)-D beta(l-6)-D glucan, beta(l-3)-D glucan, ganoderic acid and heteroglucan. In commercial standards, however, six compounds were identified in mycelium and nine compounds were found in the fruit body preparation.
