Detection of Papaya ring spot virus through double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) in Assam.

Papaya ring spot virus (PRSV) disease is a major limiting factor for commercial cultivation of papaya. The PRSV disease survey conducted in 15 districts of Assam (Jorhat, Sivasagar, Dibrugarh, Golaghat, Kamrup, Goalpara, Kokrajhar, Barpeta, Nagaon, Morigaon, Sonitpur, Darang, Udalguri, Lakhimpur and North Cacher Hills) revealed the prevalence of PRSV in all the papaya orchards. A total of 426 diseases samples were collected from these districts. The characteristic symptoms, such as mosaic, shoe string, green island, blistering, water-soaked streaking of the petioles and stems and ringspots on fruits were observed in papaya. The serodiagnostic assay DAS-ELISA was used to detect PRSV in the infected samples. The results showed that of the 426 samples, 240 samples (56.3%) were positive. The high incidence (74.3%) was noticed in Goalpara district followed by 70 per cent in Kokrajhar district (70.0%) and the lowest in Nagaon district (28.0%). The results further indicated that the highest mean PRSV titre values for all positive samples were maximum in Goalpara district (1.317 nm) followed by Barpeta district (0.708 nm), while it was lowest in Nagaon district (0.118 nm).
