A sensitive, reliable and reproducible HPLC assay for the determination of levofloxacin (LVFX) in growth media, human BAL fluid, alveolar macrophages and plasma is described. Gatifloxacin was used as the internal standard. The compounds were separated on a reverse phase column with a mobile phase consisting of acetonitrile and buffer in a ratio of 13/87, v/v, using a programmable fluorescence detector. The intra- and inter-day recovery, precision and linearity were eval-uated from all runs in all matrixes. The relative standard deviations (RSD %) of intra- and inter-day for LVFX in growth media and the three human matrixes were less than 3%. These analytical methods were successfully applied to both human samples derived from an in vivo study and growth media samples obtained during an in vitro study conducted in our laboratory.
