Response to 'physiologically based pharmacokinetic modeling at the extremes of age'

To the Editor: We are grateful to Johnston et al. for their letter, "Physiologically Based Pharmacokinetic Modeling at the Extremes of Age."We agree that there are many similarities with regard to pharmacotherapy in pediatrics and geriatrics, and that an opportunity is offered by applying physiologically based pharmacokinetic (PBPK) models in research and development for the treatment of vulnerable populations, with the ability to anticipate the impact of age, genetics, disease, and drug formulation.
