Physiologically based pharmacokinetic modeling at the extremes of age

To the Editor: We read with interest the excellent review, "Physiologically Based Pharmacokinetic (PBPK) Modeling in Children," by Barrett et al. on the use of physiologically based pharmacokinetic modeling in the pediatric population. The use of this methodology for drug development and dosing recommendations is integral to the safe arid efficacious use of medicines in vulnerable populations. The practical, logistical, and ethical challenges and methods described for conducting studies in children are strikingly similar to those for frail older adults.
