Based on the characteristic that there is crossing-over in male Bombyx mori but no crossing-over in female Bombyx mori, Dong34, a thermotolerant strain, and Ou17, a highly susceptible strain, were used to produce two backcross populations, namely, (Ou17 X Dong34)female X Ou17 male and Ou17 female X (Ou17 X Dong34)male, which were designated as BC1F and BC1M, respectively. With 84 specific primers for molecular markers selected from published SSR marker-based molecular linkage map, twenty two BC IF individuals were used to screen simple sequence repeat (SSR) molecular markers linked to thermotolerance gene (KN), and 5 SSR molecular markers on the 8(th) linkage group were found to link to KN. Subsequently, 192 BC I M individuals were used to map these markers and KN, and a linkage map was constructed, where the order of the five SSR molecular markers and KN are LFL1123, LFL0407, KN, LFL0329, LFL0658 and LFL0944.
