In Vitro Induction of Adipose-Derived Mesenchymal Stem Cells Differentiated Into Sweat Gland Epithelial Cells

Objective: In order to provide basis for the in vitro construction of eccrine sweat gland tissue, we induced the adipose-derived mesenchymal stem cells (ADSCs) to differentiate into sweat gland epithelial cells (SGECs). Methods: ADSCs were isolated and cultured in vitro. The expressions of CD29, CD34, CD45, CD90 and CD105 were detected by flow cytometry. ADSCs were induced to differentiate into osteocytes and lipocytes by osteogenic and adipogenic liquid respectively. The obtained osteocytes and lipocytes were identified by alkaline phosphatase and oil red O staining. ADSCs cultured to the third generation were treated with DMEM (10% fetal calf serum and 40 ng/ml hepatocyte growth factor (HGF)) and induced to differentiate into SGECs in matrigel. Results: The cultured ADSCs proliferated well with fusiform shape. The detection results by flow cytometry indicated that the positive rates of CD29, CD90, CD105, CD34 and CD45 were 86.4%, 97.7%, 89.8%, 3.7% and 2.3%, respectively. The induced cells presented the phenotypic characteristics of the lipocytes and osteocytes after directed induction. The characteristics of SGECs can be observed in Matrigel 5 days after treated by 40 ng/mL HGF. Conclusion: ADSCs can differentiate into SGECs which shows the possibility of constructing sweat gland tissue in vitro.
