Modulation of Process Parameters for Enhanced Production of Yellow Laccase and Its Application in Lignin Degradation

"Yellow laccases" a class of extracellular oxidoreductase enzymes oxidize the phenolic groups in lignin, dyes and many nonphenolic compounds. In the present study, response surface methodology based on central composite design has been employed for modelling of yellow laccase production from newly isolated Lentinus squarrosulus (MR13). In order to achieve further refinement of the optimized conditions for enzyme production, response surface methodology model has been integrated with Genetic algorithm. By employing Genetic algorithm, laccase production has been increased up to 26,934 IU gds~(-1) when rice straw was used as substrate. Lignin degrading ability of the crude yellow laccase has been studied on Ricinus communis, a potential lignocellulosic substrate for biofuel production. Yellow laccase was found to degrade 83.17% lignin of Ricinus communis after 7 h incubation. The enzyme is important as it can be used by industry to process lignocellulose for production of biofuel as well as other commercial products. This is the first report on enhanced production of yellow laccase from Lentinus squarrosulus (MR13) by statistical optimization.
