Transcription factors play key roles in differentiation and development as well as in controlling tissues homeostasis. It has been also firmly established that overex-pression of proliferation-associated transcription factors can lead to tumorigenesis. The strategic positions for transcription factors in gene regulation and comparatively solid understanding of their function makes these proteins highly attractive leads in cancer drug discovery; however, transcription factors have been historically considered poorly druggable. Hence, identification of small molecule inhibitors that inhibit (directly or indirectly) transcrip-tional regulators as well as mechanistic understanding of their inhibitory action attract a great deal of interest in cancer research community.
