Facile Preparation of Anticancer Drug Encapsulated Chitosan Nanospheres and Their pH Stimulus Responsive Release Property

Monodisperse chitosan nanospheres encapsulated with doxorubicin (Dox) in uniform size of similar to 400 nm were prepared by a facile ultrasound-assisted precipitation method in water. The preparation of chitosan nanospheres (Cts NSs) and the encapsulation of Dox were facilitated in one step. The drug loading/release studies reveal that these Dox@Cts NSs have a drug loading efficiency (DLE) of 78% and drug loading content (DLC) of 20 wt%. Besides, the release of Dox from Dox@Cts NSs was pH depended. At pH = 7.4, only a small amount (similar to 12%) of Dox was released. While at pH = 5.0, nearly all amount of incorporated Dox was released. These results suggest the Dox@Cts nanospheres can be prospectively used to utilize pH-responsive delivery of anticancer drug.
