This study was conducted to establish salix (Salix humboldtiana) in vitro through the induction and formation of friable callus of leaf explants to provide future regeneration of plant for the mass propagation of the species. Leaf explants were inoculated in MS medium (Murashige and Skoog, 1962) supplemented with different concentrations of 2,4-D (0, 1.0, 2.0, 4.0, 6.0, 8.0 and 12.0 mg/litre) and combinations of NAA and 6-benzylaminopurine [benzyladenine] both with the concentrations of 0, 1.0, 2.0, 4.0, 6.0 and 8.0 mg/litre. Results showed that explants inoculated with the absence of growth regulators had no formation of friable callus. Significant production of friable callus (90%) was obtained with 6.0 mg 2,4-D/litre. Individual concentrations of NAA or 6-benzyladenine induced callogenesis and rooting.
