Resistance breeding requires efficient screening methods In this paper, investigations are described which verify that tissue print-immunoblotting is suitable for routine detection of SCMV or MDMV in maize plants. In our trials, the virus detection by means of TPIB was as reliable as a simultaneous variant of a DAS-ELISA. In case of mosaic symptoms in form of single stripes on the leaf, it is absolutely necessary to take samples from this tissue, because normal green coloured parts of the leaf do not contain any virus. The stems of the maize plants are suitable for resistance screenings in the greenhouse, whereas the midrib of the leaves was used under field conditions. Because TPIB provided reliable results under extreme environment conditions (high temperatures, mechanical stress of the membranes), this method is recommended for a virus routine detection both for greenhouse and field trials. Dependent of the low demand of laboratory equipment and the low labour input, the TPIB is a low-cost alternative to the conventional ELISA.
