摘要

Distribution and localization of cherry leaf roll nepovirus (CLRV) in leaves of deciduous forest trees and herbaceous hosts revealing chlorotic spots, line patterns and chlorotic areas, were investigated by tissue print immunopressblotting (TPI). Theroutine preparation method for rolled leaf blades had to be modified for detection of virus distribution patterns in petals. This was achieved by mechanical removal of the lower epidermis and press blotting the leaves onto nitrocellulose in a vise. In Betula pendula- Roth and Prunus avium L.-leaves, detection of virus antigen generally was confined to chlorotic spots and line patterns Virus distribution in herbaceous hosts varied depending on host plant species: In leaves of Nicotiana tabatum L, cv, 'Xanthi-nc', viral antigen could only be detected in chlorotic or neciotic patterns as described for woody plants, whereas in Chenopodium quinoa Willd.-leaves, virus was also present in green areas. The results were confirmed by ELISA.
