摘要

The optimal conditions for callus induction and shoot regeneration from hypocotyls and cotyledons of sesame (S. indicum) were determined. After 4 weeks, the most effective concentrations of benzyladenine (BA) for callus induction (84.5-89.1% from the cotyledon and 85.9-89.6% from the hypocotyl) were 1 and 3 mg/litre, regardless of the concentrations of NAA. The AgNO3 concentrations most effective for callus induction (78.0-85.0% from the cotyledon and 79.0-85.6% from the hypocotyl) were 5, 10 and 15μM. Shoot regeneration from hypocotyls (25.8%) and cotyledons (10.0%) were highest in MS medium containing 3.0 mg BA/litre, 0.5 mg NAA/litre, 30 g sucrose/litre and 8 g agar/litre with AgNO3. Shoot regeneration from hypocotyls (20%) and cotyledons (8%)was promoted by AgNO3 treatment at 15μM. Treatments with 2,4-D were not effective for callus induction and shoot regeneration.
