Localization of the mRNA of early genes along cytoskeleton

USING electron microscope, cytochemical and biochemical techniques we found that parts of polysomes, some mRNAs, several initiation factors and the elongation factor 2 are bound to cytoskeleton. This means that the cytoskeleton may also play a role ingene expression. The c-jun, c-fos and c-myc proteins are transcription factors, which play an important role in the regulation of progression from Go to S phase. The disruption of the microfilaments (MF) by cytochalasin D (CD) in HeLa cells results in the inhanced transcription of the c-fos gene. In interphase HeLa cells approximately 80% of the c-fos mRNA is associated with the cytoskeletal structure. The CD treatment efficiently released the c-fos mRNA from the cy-toskeletal framework. Greater than 70% of the c-fos mRNA accumulated in the soluble phase in CD-treated cells. However, there is still no report about whether MF disruption agent has an effect on transcription of c-jun, c-myc and the c-jun, c-myc mRNA also bound to the cytoskeletal framework. We have therefore directed our efforts towards studying the effects of cytochalasin B (CB) treatment of mouse C_3H_10T_1/2 fibroblasts on the localization of c-jun, c-fos and c-myc mRNAs and the transcriptional level of these genes.
