Follicle fluid meiosis activating sterol (FF-MAS) has been described as one of the compounds inducing oocyte maturation within the follicle (Nature 1995;374:559-62). After in vivo and in vitro follicle growth, synthetic FF-MAS has been described to induce meiosis in denuded (DE) oocytes (Biol Reprod 1999;61:1362-72, J Reprod Fertil Abstract Series 2000,26:11). The role of MAS in follicle-enclosed oocyte maturation has been scarcely investigated and, therefore, in vitro studies are needed. In a dose-effect experiment using cumulus oocyte complexes (COCs) obtained after in vitro follicle culture, FF-MAS could initiate meiosis, as defined by germinal-vesicle breakdown (GVBD), at all concentrations tested (10-30 mum) and significantly stimulated PB extrusion at a dose of 30 pm. The aim of our study was to evaluate the efficacy of a FF-MAS-Agonist on meiosis induction in mouse oocytes grown from preantral follicle culture.
