Comparative Analysis of Serum Proteome in Healthy and Brucella abortus-Infected Dairy Cows

Conventional two-dimensional electrophoresis (2-DE) was used to study the serum protein changes and the value of disease-related proteins in dairy cows infected with Brucella abortus. High-abundance proteins in serum were depleted using Albumin & IgG Depletion Spintrap (TM) and proteins were retrieved by 2-D clear-up kit. Differentially expressed proteins were detected by PDQUest8.0 software and subjected to ion trap mass spectrometry equipped with a high performance liquid chromatography system. The results indicated that high-abundance protein components to be removed thoroughly were confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) combined with 2-DE gel electrophoresis. Serotransferrin, serum haptoglobin, serum amyloid A, protein HP-25 homolog 1 and apolipoprotein C-III were identified successfully from 14 differential protein spots. These proteins are mainly involved in material transport, energy metabolism, stress response and body defense. The study helps provide useful information for understanding the host immune system response for defense against pathogens at the protein level. These identified novel proteins are likely to be potential candidate targets for the development and manufacture of new vaccines.
