Partially purified xylanase from the Antarctic Paenibacillus sp. KIJ1 was non-covalently immobilized on Eudragit L-100 reversible soluble-insoluble enteric polymer. A maximal immobilization efficiency of 0.87 was obtained with final substrate concentration of 1.8% xylan at pH 6.0 and 50 degrees C. The optimum pH of the immobilized and free KIJ1 xylanase was 5.5 and 8.0-8.5, respectively, whereas both free and immobilized enzymes had an optimum temperature of activity at 50-60 degrees C. The immobilized KIJ1 xylanase could be repeatedly used in three consecutive cycles where the preparation was assayed at 50 degrees C with 1.8% xylan in 50 mM sodium phosphate (pH 6.0). The immobilized KIJ1 xylanase showed good storage stability at 25 degrees C, retaining 60% of its initial activity even after 60 d at this temperature, while the free enzyme activity declined to 40%. Immobilization of xylanase from Paenibacillus sp. KIJ1 on Eudragit L-100 may be a potential tool to address the nutritional disorder of birds caused by nonstarch polysaccharide in feed.
