Pathogenic Leptospira of the serogroup pomona were examined after being spiked in bovine semen (at concentrations from 101 to 105) samples using PCR. DNA from the spiked Leptospira were extracted using three different methods namely boiling method, lysis method and cetyl trimethyl ammonium bromide (CTAB) method. After extraction results showed that the lysis method of DNA extraction and amplification was observed to be the most sensitive as it could detect 102 Leptospira organisms spiked in the bovine semen. As for CTAB detection was attained at a concentration of 103 of Leptospira while the boiling method was not able to detect any amplicons.
