258 tissue samples collected from poultry during field outbreaks of Newcastle disease (ND) in India were examined to compare the efficacy of 5 diagnostic tests. 180 samples were positive by haemagglutination and haemagglutination inhibition test (standard HA and HI tests, 69.76%), 165 samples by latex agglutination test (LAT, 63.95%) 175 samples by avidin biotin-dot ELISA (AB-Dot-ELISA, 67.8%) and 157 samples by immunoperoxidase test (IPT, 60.85%). The sensitivity and specificity were 98.28% and 92.86% for Dot-ELISA, 97.63% and 87.64% for LAT, 99.4% and 95.12% for AB-Dot-ELISA and 95.12% and 84.78% for IPT. The only significant difference in results was between those of IPT and HA and HI tests. Dot-ELISA and LAT are recommended for diagnosing ND in field conditions because they are less costly than AB-Dot-ELISA.
