Enzyme linked immunosorbent assay (ELISA) for the detection of bovine paratuberculosis using group specific polypeptides.

Sonicated antigen of Mycobacterium paratuberculosis when analysed by SDS-PAGE revealed 36 polypeptides in the molecular weight range of 15.8 to 95.0 kD. These polypeptide bands were grouped into 4 groups based on molecular weight, cut separately, eluted and injected into rabbits for the production of antisera. This group specific antisera was used in ELISA as catching antibody and was tested against M. paratuberculosis, M. phlei and C. renale sonicate antigens. Group II and III polypeptides of M. paratuberculosis, group III in case of M. phlei and group II in case of C. renale revealed distinct positive reaction.
