RAPD based identification of farmers' collections of Kaiartamak and traditional varieties of basmati rice

A total of 18 accessions of rice (Oryza sativa L.) were subjected to random amplified polymorphic DNA (RAPD) analysis using polymerase chain reaction with nine arbitrary decamer oligonucleotide primers. Rice material studied includes three traditionalbasmati varieties (Ranbir basmati, Type-3 and Tarori basmati), six evolved basmati varieties (Pusa basmati -1, Haryana basmati-1, Pusa 1121, Super basmati, CSR 30 and Basmati -385), five strains of Kala Namak, a premium specialty scented rice of EasternUttar Pradesh, Ramkajeera. a local strain of specialty rice, NDR 359 and Satthi varieties of coarse rice and a newly developed genotype of quality rice namely Vailabh basmati 21. A total of 61 different marker bands, were observed out of which 72.13% were polymorphic. The size range of the amplified DNAs was between 300 bp to 3500 bp. The RAPD data was analyzed to generate Jaccard's coefficients for RAPD bands and the experimental material was further classified following dendrogram analysis using theunweighted pair group method with arithmetic averages (UPGMA). Results showed that Kala Namak collections expressed maximum diversity from evolved and traditional basmati varieties. The newly developed variety Vailabh basmati 21 expressed closeness withtraditional varieties of basmati rices.
