A technique for random amplified polymorphic DNA polymerase chain reaction using DNA from single aphid embryos is described. The usefulness and reliability of single-embryo analysis was evaluated using Acyrthosiphon pisum, Aphis fabae, A. frangulaeand A. gossypii. The results enabled the separation of closely related aphid species. Primer annealing at complementary locations throughout the genome allowed the synthesis of multiple PCR amplification products in all aphids tested. Embryos from the same colonies produced similar DNA patterns, regardless of whether they originated from the same or different mothers, whereas DNA extracted from embryos of different species each gave unique banding patterns. These patterns allowed the rapid differentiation of aphid populations on the basis of single individuals.
