Chromosomal DNA replication is a fundamental part of the cell division cycle and is central to life. It must be carefully regulated to ensure precise and error-free duplication of the genome, and must occur only when it is appropriate. The control of chromosomal DNA replication largely centers on the sequential deposition of initiation proteins at replication origins. Over the last few years, much has been learned about how these process are regulated, but much more remains to be discovered. This review focuses on the unanswered questions about how the initiation of replication is controlled and where research might lead in the coming years.
