For severe total limbal stem cell deficiency, the most recent treatment involves the use of cultivated epithelial stem cell transplantation. This is the latest technology for reconstructing damaged ocular surfaces caused by stem cell deficiencies. Cultivated conjunctival epithelial cells transplantation has the advantage of reduced risk of graft rejection and transmission of infection and reduces the need for long-term steroids or immunosuppression in bilateral severe cases of ocular surface disorders. We describe the surgical technique for ocular surface reconstruction using cultured conjunctival epithelial transplantation cultivated ex vivo on denuded amniotic membrane.
