rs 1800497 is a single nucleotide polymorphism (SNP), which creates and destroys the TaqA1 restriction fragment length polymorphism near the DRD2 and ANKK1 genes. The TaqAl polymorphism has been found to be both linked (Cook et al, 1996) and associated (Ponce et al, 2008) with alcoholism, rs 1800497 is responsible for an amino acid substitution within the 11th ankyrin repeat of ANKK1, which may affect the substrate-binding specificity of the ANKK1 protein (Neville et al., 2004). rsl800497 has been associated with a reduced number of dopamine-binding sites in the brain as well, which may mediate increased alcohol consumption (Blum et al, 1996).
