We read with great interest the article by Filges et al. entitled 'aCGH on chorionic villi mirrors the complexity of fetoplacental mosaicism in prenatal diagnosis' (Filges et al., 2011). The authors report possible difficulties in interpreting results when prenatal diagnosis is performed after chorionic villus sampling using an array-comparative genomic hybridization (CGH) molecular cytogenetic technique. In our Fetal Medicine Center, we have carried out more than 25 000 chorionic villus samplings since 1983 (Saura et al, 2010a). To ensure reliability in cytogenetic examinations, we have 'campaigned' actively for several years now for cytogenetic techniques to be performed not on direct chorionic villi or on the cells that make up their cytotrophoblast but on the mesenchymal core of the villi (Toutain et al, 2008, 2010a,b; Saura et al., 2010b). In our opinion, the discrepancy in results described by Filges et al. when using an array-CGH technique after DNA extraction from direct chorionic villi and after DNA extraction from cultured villi merely 'proves' this concept and justifies our concerns (Toutain et al, 2010a; Saura et al, 2010b). We would, nevertheless, like to make a few additional comments concerning the article by Filges et al.
