Detection of a specific fungal pathotype (F. o. f.sp ciceris race 5) was achieved using molecular techniques. By cloning and sequencing a 1.6 kb fragment of random amplified DNA, specific polymerase chain reaction primers were designed. Genomic DNAextracts from symptomless chickpea (cultivars PV13, PV60, CPS1 and ICCV2) plants were screened for the presence of the fungus. The polymerase chain reaction primers could detect the presence of wilt-inducing DNA in infected but symptomless plants. Blindtrials were carried out using the primers, which correctly identified the wilt inducing isolates of F. o. f.sp. ciceris. The primers could successfully discriminate between a F. o. f.sp. ciceris which caused wilt and a race which caused yellowing.
