Anesthetic effect of mixture of tanquilizer and analgesic formula an alternative to Ketamine

Ketamine that is a general anesthetic, has been popularly used to anesthetize mice for animal experiment. Because it has the tranquilized effect and analgesic effect, it is an extremely useful anesthetic used widely by using together with another anesthetic such as Xylazine. However, Ketamine is specified for the narcotic drug due to the abuse problem on January 1, 2007 by revision of" Law that manages narcotic drug and psychotropic drug ". The narcotic handler should acquire the license when Ketamine is used. It is hoped that the development of the drug to achieve the same effect of Ketamine without acquiring the narcotic license. Here, we report an injectable formula with the equivalent effect of Ketamine as an alternative to Ketamine. The formula examined was the mixture (Med/Mid/But) of sedatives, Medetomidine (Med), Midazolam (Mid), and a non-narcotic analgesic, Butorphanol (But). The ICR(7-9W) were administered with Med/Mid/But i.p., and examined them for every five minutes. The foreleg, hind leg and tail pinch reflexes and eye reflex were observed. The respiratory rate was also counted. When Med/Mid/But 0.3/4.0/5.0 was administered, the anesthetic duration was more than 30 minutes. The induction and awaking times were almost identical to those with Ketamine and Xylazine mixture. Because the anesthetic effect of the formula (Med/Mid/But) 0.3/4.0/5.0 is more than that of Ketamine and Xylazine mixture, it is thought that the formula can be used as an alternative to Ketamine in rodent anesthesia.
