Neuralcrestcell(NCC)invasionisacomplexsculptingofindividualcellsintoorganizedmigratorystreamsthatleadtoorgandevelopmentalongthevertebrateaxis.KeytoourunderstandingofhowmolecularmechanismsmodulatetheNCCmigratorypatternisinformationaboutcellbehaviors,yetithasbeenchallengingtoselectivelymarkandanalyzemigratoryNCCsinalivingembryo.Here,weapplyaninnovativeinvivostrategytoinvestigatechickNCCbehaviorswithintherhombomere4(r4)migratorystreambycombiningphotoactivationofKikGRandconfocaltime-lapseanalysisofH2B-mRFP1transfectedNCCs.Wefindthatthespatialorderofr4NCCemergencetranslatesintoadistal-to-proximalinvasionofthe2ndbranchialarch.LeadandtrailingNCCsdisplaysimilaraveragecellspeedsanddirectionalities.Surprisingly,wefindthatleadNCCsproliferatealongthemigratoryrouteandgrowtooutnumbertrailingNCCsbynearly3to1.Asimple,cell-basedcomputationalmodelreproducesther4NCCmigratorypatternandpredictstheinvasionordercanbedisruptedbyslower,lessdirectionalleadcellsorbyenvironmentalnoise.OurresultssuggestamodelinwhichNCCbehaviorsmaintainaspatially-orderedinvasionofthebranchialarcheswithdifferencesincellproliferationbetweenthemigratoryfrontandtrailingNCCs./p/div
