Inthedevelopingspinalcord,signalsoftheroofplatepatternthedorsalprogenitordomainandcontrolthespecificationofthreeneurontypes,dorsalinterneuronsdI1,dI2,anddI3.BmpandWnt/β-cateninsignalsaswellastranscriptionfactorslikeOlig3orNgn1/2areessentialinthisprocess.WehavestudiedtheepistaticrelationshipbetweenBmpandWnt/β-cateninsignalsandthetranscriptionfactorOlig3indorsalspinalcordpatterning.Usingemβ-catenin/emgain-of-functionandcompoundemβ-catenin/emgain-of-function/emOlig3/emloss-of-functionmutationsinmice,wecouldshowthatWnt/β-cateninsignalsactupstreamofemOlig3/eminthespecificationofdI2anddI3neurons.TheanalysisofsuchcompoundmutantmiceallowedustodistinguishbetweenthetwofunctionsofWnt/β-cateninsignalinginproliferationandpatterningofdorsalprogenitors.Usingelectroporationofchickspinalcords,wefurtherdemonstratethatBmpsignalsactupstreamofWnt/β-cateninintheregulationofemOlig3/emandthatWnt/β-cateninsignalsplayaninstructiveroleincontrollingemOlig3/emexpression.WeconcludethatWnt/β-cateninandBMPsignalscoordinatelycontrolthespecificationofdorsalneuronsinthespinalcord./p/div
