Brassinosteroids(BRs)playpivotalrolesinplantcelldivisionandcellelongation.Althoughboron(B)isanessentialelementforplants,excessBistoxic.Inthisstudy,weinvestigatedtheeffectsof24-Epibrassinolide(EBL)onexpressionofcellcycle(CycD3;1)andcellwall-related(TCH4andKOR)genesinleafandroottissuesofArabidopsisthaliana(L.)Heynhseedlingsunderhighconcentrationsofboricacid(BA)usingsemi-quantitativereversetranscriptase–polymerasechainreaction(RT-PCR).A.thalianaseedlingsweregrowninahydroponiccultureforfourweeks,andthentransferredtoliquidmediacontainingBA(50and100mgL-1)and/orEBL(0.01and1μM)andincubatedfor60hours.Inroottissues,BAtreatmentsdecreasedtheexpressionofCycD3;1gene,whereas0.01μMEBLtreatmentsincreasedtheexpressionlevelofthisgenewhencomparedwithplantsexposedtoBAalone.1μMEBLaloneorincombinationwithBAtreatmentssignificantlyledtodownregulationofCycD3;1gene.Ontheotherhand,applicationof1μMEBLresultedinsignificantincreaseofmRNAlevelsofKORandTCH4genesinroottissues.Inleaftissues,nosignificantdifferenceswereobservedintheexpressionlevelsofCycD3;1andKORgenesasaresultofalltreatments.BAtreatmentsincreasedtheexpressionlevelsofTCH4genesignificantlywhencomparedtocontrolinleaftissues.Toconclude,theresultsshowedthatEBLingeneralpositivelyregulatetheexpressionofcellcycleandcell-wallrelatedgenesunderhighBstressandstress-freeconditions.
