L-DOPA(L-3,4-dihydroxyphenylalanine)iscommonlyusedinthetreatmentofParkinson'sdisease.MonitoringtheconcentrationofL-DOPAinhumanplasmawillenabledoseoptimization,butisrarelyperformedbecausecurrentquantificationmethodsaretediousandtime-consuming.Inthisstudy,asimplemethodforthedeterminationofL-DOPAintheplasmaofpatientswithParkinson'sdiseasewasdeveloped.Amonolithicsilicadisk-packedspincolumnwithaphenylboronatemoiety,whichformsstableanioniccomplexeswiththecis-hydroxylgroupsofL-DOPA,wasusedtoextractL-DOPAfromplasmawithextractionrecoveriesexceeding90%.TheextractedL-DOPAwasthenseparatedonareversed-phasecolumnanddetectedelectrochemicallywithaboron-dopeddiamondelectrode.Themethod,whichhasalimitofdetectionof10fmol,wasthensuccessfullyappliedforthedeterminationofL-DOPAintheplasmaofhealthyvolunteersandpatientswithParkinson'sdisease...
