摘要

A novel application of capillary electrophoresis withnlaser-induced fluorescence detection (CE-LIF) was proposednto efficiently detect and monitor the interactionnbetween polymeric nanoparticles and the u0001-Amyloidnpeptide (Au00011-42), a biomarker for Alzheimer’s Diseasen(AD), at concentrations close to physiological conditions.nThe CE-LIF method allowed the interactionnbetween PEGylated poly(alkyl cyanoacrylate) nanoparticlesn(NPs) and the soluble Au00011-42 peptide monomersnto be highlighted. These results were confirmed bynsurface plasmon resonance (SPR) and confocal lasernscanning microscopy (CLSM). Whereas SPR showednan interaction between the NPs and the Au00011-42 peptide,nCLSM allowed the formation of large aggregatesassemblies at high NP and peptide concentrations tonbe visualized. All these results suggested that thesennanoparticles could bind the Au00011-42 peptide and influencenits aggregation kinetics. Interestingly, the non-nPEGylated poly(alkyl cyanoacrylate) NPs did not alternthe aggregation kinetics of the Au00011-42 peptide, thusnemphasizing the high level of discrimination of the CELIFnmethod with respect to NPs.
