Optical probes are essential for fluorescence-based biosensors and biomolecular quantification techniques. The most widely used approach for preparing such optical reporters involves bioconjugation, where an emissive dye is covalently bound to a probe molecule responsible for target recognition, and therefore the specificity of the assay. Proper management of reactions conditions and purification protocols are required to obtain high yields and to increase sensitivity, respectively. There is also growing interest in developing biological assays independent of traditional lock-and-key principles. These approaches circumvent the need to screen and develop specific probe molecules, such as antibodies or aptamers.
